The medical school is established under the Centrally Sponsored Scheme for 'Establishment of new (58 colleges) Medical Colleges attached with existing district/referral hospitals'.

The then Government of Madras issued an order  on June 22, 1946 that Guntur Medical College would start functioning from July1, 1946 admitting 50 students. The help rendered by Mrs. Achanta Rukmini Lakshmipathi, the then Health Minister was immense and all those who receive their education in this great institution are really indebted to her. Dr. G.V.Satynarayana Murthy took charge as the Special Officer, GMC Scheme on september 17, 1946.

Dr S P Rao, Professor in Preventive & Social Medicine is in possession of post graduate degrees in Community Medicine and Hospital Administration.. He is qualified in Epidemiology, Computer technology and Infectious diseases. Due to his penchant for field epidemiology work, he travelled Kerala, Karnataka, Andhra Pradesh, Tamil Nadu, Madhya Pradesh, Chhattisgarh and Delhi. He has been consulting epidemiologist for WHO centre for Social Science and DANIDA projects in MP.

The institution and its teaching hospital are located in a sprawling green campus located right on the Chennai-Kolkata National Highway (NH 16).  It is 23 km from the Vijayawada city the biggest Railway Junction in South India and just 5km away from Vijayawada Airport and everyday air flights are available from Hyderabad, Bangalore, Chennai and New Delhi cities.  The campus is well connected by road by Chennai, Kolkata highway number.  The Institute and Hospital was established in Rural Area in Krishna District covering the population around 30 lakhs.
